    Living with Mystery: Virtue, Truth, and Practice

    This paper examines how a person’s life may be shaped by living with a sense of the mystery of reality. What virtues, if any, are encouraged by such a sense? The first section rehearses a radical ”doctrine of mystery’, according to which reality as it anyway is, independently of human perspectives, is ineffable. It is then argued that a sense of mystery may provide ”measure’ for human lives. For it is possible for a life to be ”consonant’ with this sense -- through exercising humility, for example -- and even to emulate mystery. A further section corrects a misunderstanding about the connection between a sense of mystery and the virtues it invites, while a final section considers the relationship between living with mystery and religious faith

    Conductance fingerprint of Majorana fermions in the topological Kondo effect

    We consider an interacting nanowire/superconductor heterostructure attached to metallic leads. The device is described by an unusual low-energy model involving spin-1 conduction electrons coupled to a nonlocal spin-1/2 Kondo impurity built from Majorana fermions. The topological origin of the resulting Kondo effect is manifest in distinctive non-Fermi-liquid (NFL) behavior, and the existence of Majorana fermions in the device is demonstrated unambiguously by distinctive conductance lineshapes. We study the physics of the model in detail, using the numerical renormalization group, perturbative scaling and abelian bosonization. In particular, we calculate the full scaling curves for the differential conductance in AC and DC fields, onto which experimental data should collapse. Scattering t-matrices and thermodynamic quantities are also calculated, recovering asymptotes from conformal field theory. We show that the NFL physics is robust to asymmetric Majorana-lead couplings, and here we uncover a duality between strong and weak coupling. The NFL behavior is understood physically in terms of competing Kondo effects. The resulting frustration is relieved by inter-Majorana coupling which generates a second crossover to a regular Fermi liquid.Comment: 17 pages, 8 figure

    The SseC translocon component in Salmonella enterica serovar Typhimurium is chaperoned by SscA

    Background: Salmonella enterica is a causative agent of foodborne gastroenteritis and the systemic disease known as typhoid fever. This bacterium uses two type three secretion systems (T3SSs) to translocate protein effectors into host cells to manipulate cellular function. Salmonella pathogenicity island (SPI)-2 encodes a T3SS required for intracellular survival of the pathogen. Genes in SPI-2 include apparatus components, secreted effectors and chaperones that bind to secreted cargo to coordinate their release from the bacterial cell. Although the effector repertoire secreted by the SPI-2 T3SS is large, only three virulence-associated chaperones have been characterized. Results: Here we report that SscA is the chaperone for the SseC translocon component. We show that SscA and SseC interact in bacterial cells and that deletion of sscA results in a loss of SseC secretion, which compromises intracellular replication and leads to a loss of competitive fitness in mice. Conclusions: This work completes the characterization of the chaperone complement within SPI-2 and identifies SscA as the chaperone for the SseC translocon

    Outer Retinal Structure in Best Vitelliform Macular Dystrophy

    Importance Demonstrating the utility of adaptive optics scanning light ophthalmoscopy (AOSLO) to assess outer retinal structure in Best vitelliform macular dystrophy (BVMD). Objective To characterize outer retinal structure in BVMD using spectral-domain optical coherence tomography (SD-OCT) and AOSLO. Design, Setting, and Participants Prospective, observational case series. Four symptomatic members of a family with BVMD with known BEST1 mutation were recruited at the Advanced Ocular Imaging Program research lab at the Medical College of Wisconsin Eye Institute, Milwaukee. Intervention Thickness of 2 outer retinal layers corresponding to photoreceptor inner and outer segments was measured using SD-OCT. Photoreceptor mosaic AOSLO images within and around visible lesions were obtained, and cone density was assessed in 2 subjects. Main Outcome and Measure Photoreceptor structure. Results Each subject was at a different stage of BVMD, with photoreceptor disruption evident by AOSLO at all stages. When comparing SD-OCT and AOSLO images from the same location, AOSLO images allowed for direct assessment of photoreceptor structure. A variable degree of retained photoreceptors was seen within all lesions. The photoreceptor mosaic immediately adjacent to visible lesions appeared contiguous and was of normal density. Fine hyperreflective structures were visualized by AOSLO, and their anatomical orientation and size were consistent with Henle fibers. Conclusions and Relevance The AOSLO findings indicate that substantial photoreceptor structure persists within active lesions, accounting for good visual acuity in these patients. Despite previous reports of diffuse photoreceptor outer segment abnormalities in BVMD, our data reveal normal photoreceptor structure in areas adjacent to clinical lesions. This study demonstrates the utility of AOSLO for understanding the spectrum of cellular changes that occur in inherited degenerations such as BVMD. Photoreceptors are often significantly affected at various stages of inherited degenerations, and these changes may not be readily apparent with current clinical imaging instrumentation
